Lux receiver (LuxR+pLux)

The Lux receiver device consists of a constitutively expressed LuxR transcription regulator (BBa_C0062) upstream of a cognate pLux promoter (BBa_R0062). LuxR is activated in the presence of a 3O-C6 AHL signal and binds to pLux to initiate transcription of a downstream coding sequence.

Usage and biology

The Lux receiver is based on the Lux quorum-sensing system from Vibrio fischerii. Quorum sensing is a naturally occurring mechanism that certain strains of bacteria use to regulate gene expression in response to their population density. These bacteria secrete autoinducer signalling molecules, such as N-acyl homoserine lactones (AHLs), that bind to transcription factors to alter gene expression.

In this case, the constitutively expressed LuxR transcriptional regulator (BBa_C0062) is activated by the binding of 3O-C6 AHL, an AHL quorum signal. The activated LuxR regulator binds to the LuxR-inducible promoter, pLux, to initiate transcription of a downstream gene.

Characterisation

We characterised the activation range of this device using a GFP reporter. The results of our characterisation experiments can be found here (BBa_K1893005)
